The OPEK kit contains medium buffers that are used to "prepare & digest" tissues. OPEK effectiveness depends upon tissue quality, tissue characteristics, enzyme concentration, media ingredients, duration of enzyme-tissue exposure, removal of enzyme, and other factors. Each step is important, as improper exposures may harm cells. Tissue heterogeneity implies that cell output variations are expected with each procedure.
